Optimal glycan density for interactions between Siglec-8 and sialyl sulfo oligosaccharides

Abstract Siglec-8, a lectin involved in the suppression of allergic responses, is considered therapeutic target for developing glycan materials that enable regulation inflammatory diseases. In such materials, “multivalent effect” essential regulating immune responses. The dramatically increases apparent affinities (avidities) glycan-lectin interactions by clustering ligand multivalently. Numerous lectin-glycan utilize this effect immunological reactions, and Siglec-8 thought to show an similar those other lectins. However, we hypothesized increasing local concentration ligands modulating density are allowing interact effectively with Siglec-8. Here, investigate density-dependent changes avidities between sialyl sulfo-oligosaccharides quartz crystal microbalance (QCM) first time. As result, it was determined has optimal (~9.2 × 10 2 molecules μm −2 ) preferred interactions. Additionally, suggests key targeting results described here will promote development ameliorate disorders as inflammation anaphylaxis.
